Edit: The first hint was the opening theme song. One verse says "obake wa gakko mo shiken mo nanimo nai!" (obake don't have school or tests or anything) and another says "obake wa shinanai byouki mo nanimo nai!" (obake dont die or get sick or anything)

Second hint was Nekomusume, one of the main characters, a bakeneko/girl

Third hint was Kitaro fights with his geta, shoots his hair like a porcupine's quills (even though technically porcupine don't shoot their quills, but you understand), and has an ocarina that transforms into a stick he can fight with.

Last hint was obvious.

Our hero's first love is now married to his older brother, so he goes away to live at school.

Here's my clue.

Witnesses are usually killed, but this one is captured, brainwashed and becomes the newest assassin. With no memory of who he is, the main character is drawn into a world of conspiracy, deceit and violence.

Since he has no memory of his past in order to cope with his new existence as a mafia assassin, the main character training his mind and body until he masks his feelings and reacts like a puppet... following orders without thinking about what he is actually doing.
